Parameter estimation for metabolics is either a top-down or bottom-up approach. The top-down approach is to estimate from the time series data, the bottom-up approach is to estimate the kinetic rates through in-vitro enzymatic assays. The bottom-up method requires more time and finances to perform the experiments. From time-series data, the parameters are determined by minimizing an objective function that measures the difference between the experimental data and the model predictions.
